Refolding Protocol | Inclusion bodies (0.3-0.5g wet weight) were dissolved in 6-10ml of solubilization buffer and incubated at room temperature for 1-2h. The protein was then dialyzed against 6M GdnHCl (pH 2.0) at 4degC. Mixed disulfide bonds were produced by the addition of an equal volume of 6M GdnHCl, 0.4M oxidised glutathione, 4mM EDTA, 0.2M Tris pH 8.0 to the denatured protein solution. After 2-3h incubation at room temperature the solution was then dialyzed against 6M GdnHCl/0.1M Tris, 2mM EDTA pH 8.0 at 4degC. The protein was then diluted to a final concentration of 20microgram/ml or less in refolding buffer. The mixture was incubated at 10degC for 3-4 days, the protein was then concentrated. |
