Expression Protocol | Cells were grown in LB broth at 37degC until OD600 reached 0.8. 0.4mM IPTG was added and cells were grown for a further 3h. Cells were then collected by centrifugation and washed in 10mM TrisHCl pH 8, 1mM EDTA, 100mM NaCl. |

Refolding Protocol | Cells were lysed by 3 cycles of rapid freeze/thaw in the presence of 0.1mg/ml lysozyme, and sonicated twice for 30sec. The cell lysate was centrifuged, and the pellet was then resuspended by sonication with a cuphorn probe into wash buffer and recentrifuged. After 3 cycles of washing, the pellet was dissolved in a small amount of 6M urea, then diluted with 9 volumes of refolding buffer 1 and then dialyzed against refolding buffer 2 then refolding buffer 3. |
