Refolding Protocol | Inclusion bodies obtained from lysed cells were suspended in solubilization buffer to give a protein concentration of 2mg/ml. After incubation at 30degC for 2h, the solution was centrifuged (10000g, 10min). The supernatant was diluted 10-fold with refolding buffer 1 and left at 15degC for 2h. The sthe solution was adjusted to 8.0 with 1N HCl, maintained for 1h and dialyzed against refolding buffer 2 at 4degC overnight. The protein was then activated at pH 2 and 15-20degC for 2-14h to produce pseudochymosin, which was then dialysed against 0.1M sodium acetate pH 5.5 at 25degC overnight to facilitate activation of the protein |
