Refolding Protocol | Denature inclusion bodies in 6M GuHCl for several days. The protein was then absorbed onto zeolite and incubated for 30mins. Following incubation the samples were centrifuged 10,000 rpm for 5 seconds. They were then washed with PBS x4 and resuspend in 50mM Hepes (pH 7.5), 0.5M NaCl, 0.5% (w/v) PEG 20,000, 20 mM beta mercaptoethanol and 1% (v/v) Tween 20. The mixture was then allowed to refold at 4 C for 10hrs. |
